Step 1. Prepare Sample & Load Hemacytometer
Pipette 20 microliters of sample into the Nexcelom disposable hemacytometer
Step 2. Manually Count Cells in Sample
a. Place the disposable hemacytometer under a microscope with a typical magnification of 100.
b. Focused both onto the grid pattern and the cell particles, and count the total number of cells found in 4 large corner squares.
Step 3. Cell Calculations & Disposal of Hemacytometer
a. Multiply the dilution factor by the total number of cells and divide by the sample volume.
b. Dispose the hemacytometer.
